A DNA tests works by comparing certain sections of DNA that are present in everyone. In the case of parents and children, because you get half your DNA from your mother and half from your father, there is a 50% of that section coming from each parent. If the sections match up at the same time, then they are a match. If they come at different times then there is a really small likelihood they are a match.
